ABSTRACT

A mucosite oral é um quadro clínico que acomete frequentemente pacientes sob terapia antineoplásica na região de cabeça e pescoço e caracteriza-se por ulcerações na mucosa que geram intensa dor local, odinofagia, aumento do risco de infecções, do uso de antibióticos e do tempo de hospitalização. A correlação entre mucosite oral, infecção fúngica e o potencial de disseminação fúngica sistêmica foi recentemente descrita. Apesar do impacto desse quadro clínico sobre a qualidade e tempo de vida dos pacientes oncológicos, não há consenso sobre a profilaxia e o protocolo terapêutico. O plasma de baixa temperatura sobre pressão atmosférica (LTAPP) apresenta efeito antimicrobiano, anti-inflamatório e reparador tecidual, o que sugere que possa ser promissor no tratamento da mucosite oral. Os objetivos gerais deste projeto foram divididos em dois subprojetos: 1) Definir os melhores parâmetros in vitro com efeito antifúngico e não tóxico e avaliar o LTAPP no tratamento de lesão de mucosite oral em modelo murino de mucosite por quimioterapia e 2) avaliar se o tratamento com LTAPP pode prevenir a disseminação fúngica sistêmica em ratos a partir de infecção experimental de lesões de mucosite oral por Candida albicans. Para tal, foram incluídos no estudo 100 ratos (Rattus norvegicus) com 90 a 100 dias de idade. No subprojeto 1, a lesão de mucosite oral foi induzida por administração de 5 fluorouracila (5-FU), enquanto no subprojeto 2, utilizou-se 5-FU associada à cisplatina ambas associadas à aplicação tópica de ácido acético 50%. Para o subprojeto 1, os animais foram randomicamente divididos em 2 grupos experimentais (n=30): a) Grupo mucosite; b) Grupo mucosite tratado com LTAPP, avaliados após 1, 5 e 12 dias do tratamento. Durante o período experimental, as lesões foram fotografadas e a gravidade da mucosite classificada por meio da atribuição de escores. Após a eutanásia e o processamento, os cortes histológicos corados por hematoxilina-eosina (HE) foram analisados microscopicamente. Para o subprojeto 2, o estudo de disseminação sistêmica fúngica nos grupos de mucosite infectada com C. albicans tratado ou não com LTAPP foi conduzido pelo isolamento fúngico a partir de amostras de sangue total e macerado dos órgãos. Para tanto foram estudados 2 grupos de ratos (n=20): c) Grupo mucosite infectado com C. albicans e d) Grupo mucosite infectado com C. albicans tratada com LTAPP, avaliados após 24 e 72 h do tratamento. Para ambos os projetos, o melhor parâmetro in vitro foi selecionado, isto é aquele com maior atividade antifúngica e baixa toxicidade. Dessa forma, as lesões foram expostas ao LTAPP de hélio por 5 min na distância de 1,5 cm na potência de 1 W. Os resultados in vitro mostraram que o LTAPP teve efeito antifúngico e baixa toxicidade para células de mamíferos. Os resultados in vivo mostraram que 5-FU afetou a saúde geral dos animais, evidenciada pela perda de peso corporal. Em ambos os grupos, houve reparo tecidual após 12 dias do tratamento, com resolução quase completa da lesão, o que foi corroborado pelos achados microscópicos. O grupo LTAPP exibiu uma tendência maior de redução da lesão, após 12 dias de tratamento. Além disso, o LTAPP apresentou efeito inibitório sobre C. albicans após 5 minutos, de exposição, com redução da recuperação fúngica da língua após 24 h (p<0.05). A disseminação fúngica sistêmica foi reduzida significativamente após 24 e 72 h do tratamento. Com base nos resultados obtidos, conclui-se que o LTAPP é uma ferramenta promissora para futura aplicação clínica em pacientes com mucosite oral. (AU)


Oral mucositis is a clinical condition that frequently affects patients undergoing antineoplastic therapy in the head and neck region and is characterized by mucosal ulcerations that generate intense local pain, odynophagia, increased risks of infections, use of antibiotics and the length of hospital stay. The correlation among oral mucositis, fungal infection and the potential for systemic fungal dissemination has recently been described. Despite the impact of this clinical condition on the quality and life expectancy of cancer patients, there is no consensus on prophylaxis and the therapeutic protocols. Low temperature atmospheric pressure plasma (LTAPP) has antimicrobial, anti-inflammatory and tissue repairing effects, which suggests that it can be promising in the treatment of oral mucositis. The general objectives of this project were divided into two subprojects: 1) Define the best antifungal and non-toxic in vitro parameters and to evaluate the application of LTAPP in the treatment of oral mucositis in murine model for chemotherapy, and 2) to evaluate whether treatment with LTAPP can prevent systemic fungal dissemination in rats from experimental infection of oral mucositis lesions by Candida albicans. A total of 100 rats (Rattus norvegicus) aged 90 to 100 days were included in the study. In subproject 1, oral mucositis lesion was induced by administration of only 5- fluorouracil (5-FU), while in subproject 2, administration and systemic administration of 5-FU associated with cisplatin, both associated with topical application of 50% acetic acid. For subproject 1, the animals were randomly divided into 2 experimental groups (n=30):a) Mucositis group and b) Mucositis group treated with LTAPP evaluated after 1, 5 and 12 days of treatment. During the experimental period, the lesions were photographed, and the severity of mucositis was classified into scores. After euthanasia and processing, the histological cuts stained by hematoxylin-eosin (HE) were analyzed. For subproject 2, the study of fungal systemic dissemination in groups of mucositis infected with C. albicans treated or not with LTAPP was conducted by fungal isolation from whole blood and macerated organs. Therefore, 2 groups of rats (n=20) were studied: c) Mucositis group infected with C. albicans and d) Mucositis group infected with C. albicans treated with LTAPP, evaluated after 24 and 72 h of treatment. For both subprojects, the best in vitro parameter was selected, that is, the one with the greatest antifungal effect and low toxicity. Thus, the lesions were exposed to helium LTAPP for 5 min at a distance of 1.5 cm at power of 1 W. In vitro results showed that LTAPP has an antifungal effect and low toxicity. In vivo results showed that 5-FU affected the general health of animals evidenced by body weight loss. In both groups, there was tissue repair after 12 days of treatment, with almost complete resolution of the lesion, which was corroborated by the microscopic findings. LTAPP group showed a greater trend of reduction of lesion, after 12 days of the treatment. Furthermore, LTAPP showed inhibitory effect on C. albicans after 5 min of exposition, with reduction in fungal recovery from the tongue after 24 h (p<0.05). Reduction in fungal dissemination was observed after 24 and 72 h of LTAPP treatment (p<0.05). Based on the obtained results, it was concluded that LTAPP is a promising tool for future clinical application in patients with oral mucositis.(AU)

ABSTRACT

Abstract Introduction: The objective of this study is to compare the effects of conventional monopolar electrosurgery (CMES) and low-thermal plasma kinetic cautery (PKC) on complications such as bleeding, abnormal wound healing, pain, and drainage in patients who underwent on-pump coronary artery bypass grafting (CABG). Methods: This retrospective clinical study included 258 patients undergoing CABG; the patients were randomized to PKC (PEAK PlasmaBlade, n=153) and CMES (n=105) groups. The patients' clinical data were examined retrospectively for biochemical variables, postoperative drainage, post-surgery erythrocyte suspension transfusion count, surgical site pain examined with visual analogue scale (VAS), and wound healing. Two-sided P-value > 0.05 was considered as statistically significant. Results: The median post-surgery erythrocyte suspension transfusion number was significantly lower with PKC compared to CMES (0 [0-1] vs. 1 [1-4], respectively, P<0.001). Mean postoperative drain output and time until removal of drain tubes were significantly lower with PKC compared to CMES (300±113 vs. 547±192 and 1.95±1.5 vs. 2.44±1.8; P<0.001 and P=0.025, respectively). Mean VAS score for spontaneous and cough-induced pain were significantly lower with PKC compared to CMES (1.98±1.51 vs. 3.94± 2.09 and 3.76±1.46 vs. 5.6±1.92; P<0.001 for both comparisons). Reoperation due to bleeding was significantly higher with CMES compared to PKC (0 vs. 11 [7.2%], P=0.001). Conclusion: Use of PKC during CABG considerably reduces postoperative drainage, need for blood transfusion, reoperation due to bleeding, and postoperative pain. PCK appears to be a good alternative to CMES for CABG.

ABSTRACT

A etiopatogenia da doença periodontal é complexa e exige estudos constantes em alternativas terapêuticas que possibilitem seu controle. Aggregatibacter actinomycetemcomitans está associado fortemente à periodontite crônica e à periodontite agressiva. A tecnologia do plasma frio sob pressão atmosférica tem potencial para uso na odontologia, mas sua aplicação na periodontia ainda é pouco explorada. Este estudo teve como objetivo avaliar os efeitos do plasma de baixa temperatura sob pressão atmosférica (PBTPA) como adjuvante no controle da doença periodontal experimental induzida por ligadura, inoculada com A. actinomycetemcomitans, na região cervical do primeiro molar inferior em modelo murino. Inicialmente, foram determinados os parâmetros físicos efetivos para inibição de biofilmes de A. actinomycetemcomitans utilizando cepa padrão American Type Culture Collection ATCC 29523. Foram realizados testes de screening inicial com a verificação da inibição do crescimento por exposição ao PBTPA de A. actinomycetemcomitans semeados em placas de Petri. Posteriormente, foram formados biofilmes em placas de microtitulação e foram determinados os parâmetros físicos efetivos do PBTPA. Foram realizados experimentos in vivo de indução de doença periodontal por A. actinomycetemcomitans em modelo utilizando ligaduras em ratos. A tomografia computadorizada de feixe cônico foi utilizada para avaliar o nível de perda óssea após período experimental. Os resultados mostraram que o PBTPA foi efetivo na eliminação do micro-organismo observado através da formação de halos de inibição nos tempos de 5 e 7, 5 minutos e sobre biofilmes de A. actinomycetemcomitans expostos ao PBTPA no tempo de 5 minutos. Os resultados exibiram diferença estatística significativa (p <0,001) na distância entre a junção esmalte-cemento e a crista óssea alveolar, do grupo tratado com PBTPA em relação ao grupo raspagem e ao grupo controle. Os resultados do presente estudo sugerem o potencial do PBTPA no controle da periodontite induzida, contudo análises adicionais são necessárias(AU)


The etiopathogenesis of periodontal disease is complex requiring constant studies of therapeutic alternatives that enable its control. Aggregatibacter actinomycetemcomitans is strongly associated with chronic periodontitis and aggressive periodontitis. Cold plasma technology under atmospheric pressure has potential for use in dentistry, but its application in periodontics is still not well explored. This study aimed to evaluate the effects of low temperature atmospheric pressure plasma (PBTPA) as an adjuvant in the control of experimental periodontal disease induced by ligature inoculated with A. actinomycetemcomitans in the cervical region of the lower 1st molar in a murine model. Initially, effective physical parameters for inhibition of A. actinomycetemcomitans biofilms were determined using the standard strain American Type Culture Collection ATCC 29523. Initial screening tests were performed to verify growth inhibition after exposure of A. actinomycetemcomitans seeded in Petri dishes to the PBTPA. Subsequently, biofilms were formed on microtiter plates and the effective physical parameters of the PBTPA were determined. In vivo experiments were carried out to induce A. actinomycetemcomitans periodontal disease induced by ligature in a rat model. Cone beam computed tomography was used to assess the level of bone loss after the experimental period. The results showed that PBTPA was effective in eliminating the microorganism observed through the formation of halos inhibition during 5 and 7.5 minutes and in A. actinomycetemcomitans biofilms exposed to PBTPA at 5 minutes. The previous study using PBTPA in 5 minutes did not show cytotoxicity in Vero cells. The results showed a statistically significant difference (p <0.0001) in the distance between the enamel-cement joint and the alveolar bone crest, of the group treated with PBTPA in relation to the scalling group and the control group, confirming the potential of the technique in dentistry(AU)

ABSTRACT

Caracterizar a superfície de zircônia (Y-TZP) submetida a diferentes tratamentos de superfície (com e sem associações de plasma) e avaliar a resistência ao cisalhamento (RC) com a cerâmica de cobertura (CC) após diferentes métodos de envelhecimento. 301 espécimes de 3Y-TZP (IPS e.max ZirCAD) foram fabricados e distribuídos em 7 grupos (n = 43): Grupo C (controle): sem tratamento; Grupo Al: jateamento com partícula de Al2O3 de 27 µm; Grupo L: liner; Grupo P: plasma; Grupo Al+L: jateamento + liner; Grupo Al+P: jateamento + plasma; Grupo P+L: plasma + liner. A superfície de Y-TZP foi caracterizada por Micrsocopia Eletrônica de Varredura (MEV), Espectroscopia por Energia Dispersiva (EED), Microscopia por Força Atômica (MFA), perfilometria de superfície e energia livre de superfície (ELS). Difração de Raio-x (DRX) foi usada para verificar a estrutura do cristal de Y-TZP após cada tratamento de superfície realizado. A RC entre Y-TZP e a cerâmica de cobertura foi verificada após três protocolos de envelhecimento: inicial, após envelhecimento hidrotérmico (autoclave por 5h) ou termociclagem (30.000 banhos - 5-55°C). ANOVA de um (profilometria e ELS) e de dois fatores (RC) e o teste de Tukey HSD foram usados como análise estatística. Para os grupos de plasma, uma cobertura homogênea e total da superfície foi observada em MEV e MFA, com formação globular. Picos de Si foram encontrados para os grupos Al, L, Al+L e P+L. A rugosidade foi menor para os grupos C, P e Al+P. Para ELS, os maiores valores foram encontrados quando o liner foi aplicado (> 74,59 nm/Nm). O maior conteúdo monoclínico foi observado para Al+L (6,96%) e Al+P (5,86%). Para o período inicial, o grupo Al e P+L apresentaram os menores valores de RC ( 0,331). Após o envelhecimento hidrotérmico, os grupos L, P e P+L apresentaram os maiores valores de RC (> 8,25 MPa; P< 0,001). Após a termociclagem, os grupos P, L, Al+L e Al+P apresentaram os maiores valores de RC (> 6,35 MPa; P < 0,001). Foram constatadas alterações na topografia da Y-TZP e na RC com a CC, de acordo com os tratamentos realizados. O tratamento com plasma melhorou a resistência ao cisalhamento e não causou transformação de fase(AU)


To characterize the zirconia (Y-TZP) surface submitted to different surface treatments (with and without plasma associations) and to evaluate the shear bond strength (SBS) with the veneering ceramic (VC) after different aging methods. 301 3Y-TZP specimens (IPS e.max ZirCAD) were fabricated and distributed into 7 groups (n=43): Group C (control): no treatment; Group Al: airborne abrasion with 27 µm Al2O3 particle; Group L: liner; Group P: plasma; Group Al+L: airborne abrasion + liner; Group Al+P: airborne abrasion + plasma; Group P + L: plasma + liner. The Y-TZP surface was characterized by SEM, EDS, AFM, surface profilometry, and surface-free energy (SFE). XRD was used to verify the crystal structure after each surface treatment performed. SBS between Y-TZP and the veneering ceramic was verified after three aging protocols: initial, after hydrothermal aging (autoclave for 5h), or thermal fatigue (30,000 baths - 5-55°C). One- (profilometry and SFE) and two-way ANOVA (SBS), and Tukey's HSD test were used as statistical analysis. For the plasma groups, a homogeneous and a full surface coverage was observed on SEM and AFM, with globular formation. Peaks of Si were found for Al, L, Al+L, and P+L groups. Roughness was lower for groups C, P, and Al+P. For SFE, the highest values were found when the liner was applied (>74.59 nm/Nm). The highest monoclinic content was observed for Al+L (6.96%) and Al+P (5.86%). For the initial period, group Al and P+L presented the lowest SBS values (< 5.85 MPa; P> 0.331). After hydrothermal aging, groups L, P, and P+L presented the highest SBS values (> 8.25 MPa; P< 0.001). After thermal fatigue, P, L, Al+L, and Al+P groups had the highest SBS values (> 6.35 MPa; P< 0.001). Changes in Y-TZP topography and the SBS with the VC were observed, according to treatments performed. Plasma treatment improved SBS and did not cause phase transformation(AU)

ABSTRACT

O manejo clínico e tratamento das feridas crônicas têm sido considerados grandes desafios nas áreas médicas. Infecções por biofilmes em feridas crônicas afetam milhões de pessoas no mundo a cada ano e muitas mortes ocorrem como consequência. O plasma de baixa temperatura sob pressão atmosférica (LTAPP) é uma mistura gasosa contendo partículas carregadas, radicais livres e radiação. As ações biológicas das espécies reativas geradas concomitantemente, em particular antimicrobiana, anti-inflamatória e indutora de reparação tecidual, mostram-se altamente promissoras dentro do contexto do controle das feridas crônicas. O objetivo deste estudo foi avaliar os efeitos LTAPP frente a biofilmes monoespécie e multiespécies de micro-organismos frequentemente isolados de feridas crônicas. Além disso, objetivou-se avaliar estudar os efeitos citotóxicos e genotóxicos do LTAPP nos parâmetros físicos eficazes para o controle dos biofilmes A seguir, o protocolo eficaz e seguro foi avaliado in vivo utilizando modelo murino e modelo de úlcera dermal em coelhos. O LTAPP foi capaz de reduzir as UFC/mL de biofilmes monoespécie e mutiespécie a partir de 5 minutos de exposição. O LTAPP, nos parâmetros efetivos antibiofilme, apresentou-se seguro para uso em ensaios in vivo, pois não foi cito ou genotóxico. O tratamento com LTAPP levou à maior redução da área da ferida quando comparado ao grupo controle, em ambos os modelos de ferida com infecção polimicrobiana. Redução da resposta inflamatória foi observada no modelo de úlcera dermal no tempo de 8 dias. Não houve evidências de que o LTAPP aumente significativamente a proliferação celular. Com base nos resultados obtidos, conclui-se que o LTAPP acelera o processo de reparação de feridas, porém o mecanismo de ação ainda precisa ser elucidado(AU)


The clinical management and treatment of chronic wounds have been considered one of the main challenges in the medical area. Biofilm infections in chronic wounds affect millions of people worldwide each year and many deaths occur as consequence. LTAPP is a gas containing charged particles, free radicals and radiation. The biological actions of the reactive species generated concomitantly, anti-inflammatory and tissue repair inducing, are highly promising within the context of chronic wound control. The aim of the present project is to evaluate the effects of low temperature plasma under atmospheric pressure (LTAPP) on mono and multispecies biofilms frequently isolated from chronic wounds. The cyto and genotoxic effects of LTAPP in the effective parameters were also assessed. The effective and safe protocol was tested in vivo using a murine model and a rabbit dermal ulcer model. LTAPP was able to reduce significantly the mono and multispecies biofilm after 5 minutes exposure. In the anti-biofilm effective parameters, LTAPP was not cyto or genotoxic. Treatment with LTAPP reduced significantly the wound area when compared to control in both animal models. Anti-inflammatory effect was observed in dermal ulcer model at 8 days. No evidences of increase in cell proliferation caused by LTAPP were noted. According to the results, it could be concluded that LTAPP enhanced the wound repair process, however the mechanism of action must be still elucidated(AU)

ABSTRACT

Este estudo avaliou o efeito do plasma frio nas características físicas (reologia e tamanho de partícula), propriedades térmicas e microestrutura de achocolatados comparados a pasteurização (controle). Bebidas tratadas por plasma frio apresentaram partículas com maior tamanho e consistência e perfil de fusão alterado (menor temperatura, água ligada e maior entalpia) do que o produto pasteurizado, sugerindo processos de desnaturação. Condições mais severas resultaram em maior consistência (partículas de maiores áreas superficiais [D 3,2] e diâmetros volumétricos [D 4,3]). Condições intermediárias resultaram em produtos com características mais semelhantes às bebidas pasteurizadas. Os resultados indicam que bebidas lácteas achocolatadas com diferentes características poderiam ser obtidas variando os parâmetros do processo de plasma frio.

ABSTRACT

Objective: to evaluate the effect of aluminum oxide sandblasting (AOS), argon plasma application (APL) and their combination (AOS+APL) on the shear bond strength (SBS) of dual-cure resin cement (Panavia F 2.0, Kuraray Noritake) to two zirconia-based ceramics (Lava, 3M ESPE and Katana, Kuraray Noritake). Material and Methods: One hundred twenty zirconia plates (13 mm in length X 5 mm in width X 1 mm in thickness) were prepared and treated according to the following treatments: 1- AOS, 2- APL,3- AOS+APL and 4- no treatment (control). After treatments, resin cement cylinders (1.4 mm in diameter x 1 mm in height) bonded to zirconia surface were obtained by filling up the silicon matrix. The shear bond test was performed following storage of the samples for 24 hours and one year. Bond strength values were recorded in MPa and the data were analyzed by three-way ANOVA and Tukey post-hoc test (preset alpha of 0.05). Results: In general, AOS and AOS +APL showed the higher SBS. After one-year storage, SBS of the resin cement to the Katana zirconia did not reduce, regardless of the zirconia surface treatment. Conclusion: AOS alone was able to increase the SBS of the resin cement to both zirconia ceramics compared to control and SBS remained stable after one year depending on type of zirconia ceramic. Objetivo: avaliar o efeito do jateamento de óxido de alumínio (AOS), aplicação de plasma de argônio (APL) e sua combinação (AOS + APL) na resistência ao cisalhamento (SBS) do cimento resinoso dual (Panavia F 2.0, Kuraray Noritake) a duas cerâmicas à base de zircônia (Lava, 3M ESPE e Katana, Kuraray Noritake). Material e Métodos: Cento e vinte placas de zircônia (13 mm de comprimento X 5 mm de largura X 1 mm de espessura) foram preparadas e tratadas de acordo com os seguintes tratamentos: 1- AOS, 2- APL, 3- AOS + APL e 4 - sem tratamento (controle). Após realizados os tratamentos, com a utilização de uma matriz de silicone (1,4 mm de diâmetro x 1 mm de altura) foram confeccionados cílindros de cimento resinoso na superfície da zircônia. O teste de cisalhamento foi realizado após o armazenamento das amostras por 24 horas e um ano. Os valores de resistência de união foram registrados em MPa e os dados foram analisados por ANOVA de três fatores e teste post-hoc de Tukey (alfa de 0,05). Resultados: Em geral, AOS e AOS + APL apresentaram a maior SBS. Após um ano de armazenamento, o SBS do cimento resinoso para a zircônia Katana não reduziu, independentemente do tratamento da superfície da zircônia. Conclusão: AOS isoladamente foi capaz de aumentar a SBS do cimento resinoso para ambas as zircônias em comparação com o controle e a SBS permaneceu estável após um ano, dependendo do tipo de zircônia. PURPOSE: Direct application of atmospheric-pressure plasma jets (APPJs) has been established as an effective method of microbial decontamination. This study aimed to investigate the bactericidal effect of direct application of an APPJ using helium gas (He-APPJ) on Porphyromonas gingivalis biofilms on sandblasted and acid-etched (SLA) titanium discs. METHODS: On the SLA discs covered by P. gingivalis biofilms, an APPJ with helium (He) as a discharge gas was applied at 3 different time intervals (0, 3, and 5 minutes). To evaluate the effect of the plasma itself, the He gas–only group was used as the control group. The bactericidal effect of the He-APPJ was determined by the number of colony-forming units. Bacterial viability was observed by confocal laser scanning microscopy (CLSM), and bacterial morphology was examined by scanning electron microscopy (SEM). RESULTS: As the plasma treatment time increased, the amount of P. gingivalis decreased, and the difference was statistically significant. In the SEM images, compared to the control group, the bacterial biofilm structure on SLA discs treated by the He-APPJ for more than 3 minutes was destroyed. In addition, the CLSM images showed consistent results. Even in sites distant from the area of direct He-APPJ exposure, decontamination effects were observed in both SEM and CLSM images. CONCLUSIONS: He-APPJ application was effective in removing P. gingivalis biofilm on SLA titanium discs in an in vitro experiment.

OBJECTIVE@#To investigate the effect of different treatment temperatures of a novel cold atmospheric plasma jet treatment on the resin-dentin bonding.@*METHODS@#(1) Fifty-two freshly extracted, non-carious and intact third molars were collected. The occlusal one-third of the crown was removed by means of a water-cooled low-speed Isomet saw. One dentin disc [(900 ±100) μm] was prepared for each tooth. The fifty-two dentin discs were randomly divided into control group and experimental groups, of which four were in control group, and forty-eight were divided into four experimental groups according to the different treatment temperatures (4 °C, 10 °C, 20 °C and 30 °C) of the novel radio-frequency atmospheric-pressure glow discharge (RF-APGD) plasma jet, twelve in each group. Each experimental group was divided into three subgroups according to different treatment time (10 s, 20 s and 30 s), with four in each subgroup. The occlusal one-third of the crown was removed by means of a water-cooled low-speed Isomet saw. The morphology of demineralized dentin surfaces was analyzed using field emission scanning electron microscopy. (2) Twenty unerupted, non-carious and intact third molars were randomly divided into five groups, four in each group: control group, untreated; 4 °C, 10 °C, 20 °C and 30 °C experimental groups, each group was treated with the RF-APGD plasma jet for 20 s. The micro-tensile resin dentin bond strength was tested after 20 s RF-APGD plasma jet treatment with different temperatures, using a universal mechanical machine.@*RESULTS@#(1) The field emission scanning electron microscopy results indicated that when compared with the control group, a 10 s RF-APGD plasma jet treatment with 30 °C and 20 °C collapsed the collagen scaffold. Collagen fibrils maintained an uncollapsed three-dimensional structure after the 4 °C RF-APGD plasma jet treatment for even 30 s treatment. (2) The microtensile resin dentin bond strength results of the 4 °C RF-APGD plasma jet treatment group (57.8±0.7) MPa were significantly higher than that of the control group [(47.4±0.5) MPa] and 10 s, 20 s and 30 s RF-APGD plasma treatment group [(51.9±0.7) MPa,(29.7±1.0) MPa and (22.2±1.5) MPa] with statistically significant difference (P<0.05). Compared with the control group, the micro-tensile bond strength increased about 21.9% and 9.5% after 4 °C and 10 °C RF-APGD plasma jet treatment, respectively.@*CONCLUSION@#Compared with other treatment temperatures, this novel RF-APGD plasma jet treatment with the temperature of 4 °C can preserve the three-dimensional morphology of demineralized dentin better, and can improve the resin-dentin bonding.

ABSTRACT

Nonthermal atmospheric plasma has been studied for its many biomedical effects, such as tooth bleaching, wound healing, and coagulation. In this study, the effects of dentinal tubules occlusion were investigated using fluoride-carboxymethyl cellulose (F-CMC) gel, nano-sized hydroxyapatite (n-HA), and nonthermal atmospheric plasma. Human dentin specimens were divided to 5 groups (group C, HA, HAF, HAP, and HAFP). Group HA was treated with n-HA, group HAF was treated with n-HA after a F-CMC gel application, group HAP was treated with n-HA after a plasma treatment and group HAFP was treated with n-HA after a plasma and F-CMC gel treatment. The occlusion of dentinal tubules was investigated using scanning electron microscopy (SEM) and energy dispersive x-ray spectroscopy (EDS), which shows Ca/P ratio. In the EDS results, a higher Ca/P ratio was shown in the groups including n-HA than in the control group. The specimens of group HAP and HAFP had a higher Ca/P ratio in retentivity. In the SEM results, there was not a significant difference in the amount of times applied. Therefore, this study suggests F-CMC gel and n-HA treatment using nonthermal atmospheric plasma will be a new treatment method for decreasing hypersensitivity.

ABSTRACT

The aim of this study was to investigate the synergistic effect of cold atmospheric plasma (CAP) treatment and RGD peptide coating for enhancing cellular attachment and proliferation over titanium (Ti) surfaces. The surface structure of CAP-treated and RGD peptide-coated Ti discs were characterized by contact angle goniometer and atomic force microscopy. The effect of such surface modification on human bone marrow derived mesenchymal stem cells (hMSCs) adhesion and proliferation was assessed by cell proliferation and DNA content assays. Besides, hMSCs' adhesion and morphology on surface modified Ti discs were observed via fluorescent and scanning electron microscopy. RGD peptide coating following CAP treatment significantly enhanced cellular adhesion and proliferation among untreated, CAP-treated and RGD peptide-coated Ti discs. The treatment of Ti surfaces with CAP may contribute to improved RGD peptide coating, which enables increased cellular integrations with the Ti surfaces.

Abstract Objective The aim of this study was to evaluate the effect of photodynamic therapy (PDT) and non-thermal plasma (NTP) on adhesion and sealer penetration in root canals. Material and Methods Sixty single-rooted premolars were used. The teeth were prepared using a crown-down technique. NaOCl and EDTA were used for irrigation and smear layer removal, respectively. The root canals were divided into three groups: control, PDT, and NTP. After treatments, the roots were filled using gutta-percha and either AH Plus (AHP) or MTA Fillapex (MTAF) sealers. Samples were sectioned at 4, 8, and 12 mm from the apex (1-mm slices)and analyzed by the push-out bond strength test (adhesion) and confocal laser scanning microscopy (sealer penetration). Data were statistically evaluated using Kruskal-Wallis, Dunn's, and Spearman's tests. Results Regarding AHP, bond strength was similar in the NTP group and in the control group, but significantly lower in the PDT group. As to MTAF, both therapies showed lower values than the control group. In the confocal analysis of AHP, maximum and mean penetration, and penetrated area were statistically higher in the control group than in the PDT and NTP groups. Penetrated perimeter was similar among groups. Regarding MTAF, all parameters yielded better results in the NTP than in the control group. The PDT and control groups showed similar results except for penetrated area. Conclusion PDT and plasma therapy affected the adhesion and sealer penetration of root canals filled with AH Plus and MTA Fillapex and there is no positive correlation between adhesion and sealer penetration.

Abstract Purpose: To compare the effectiveness of anal and perianal condylomata treatment using argon plasma and electrofulguration. Methods: From January 2013 to April 2014, 37 patients with anal and perianal condylomata, who had been diagnosed through proctological examination, oncotic cytology, polymerase chain reaction (PCR) and histology, underwent treatment with argon plasma and electrofulguration. The perianal and anal regions were divided into two semicircles. Each semicircle was treated using one of the methods by means of simple randomization. Therapeutic sessions were repeated until all clinical signs of infection by HPV were eliminated. The patients were evaluated according to several variables like the genotype of HPV, HIV infection, oncological potential per genotype, oncotic cytology and histology. Results: Among all the variables studied, only immunosuppression due to HIV influenced the results, specifically when the fulguration method was used. There was no significant difference in effectiveness between argon and fulguration based on lesion relapse (p > 0.05). However, among HIV-positive patients, fulguration presented worse results, with a significant difference (p = 0.01). Conclusion: Regarding treatment of anal and perianal condylomata acuminata, comparison between applying fulguration and argon demonstrated that these methods were equivalent, but use of fulguration presented more relapses among HIV-positive patients.

ABSTRACT

PURPOSE: The fourth state of matter, plasma is known as an ionized gas with electrons, radicals and ions. The use of non-thermal plasma (NTP) in cancer research became possible because of the progresses in plasma medicine. Previous studies on the potential NTP-mediated cancer therapy have mainly concentrated on cancer cell apoptosis. In the present study, we compared the inhibitory effect of NTP on cell migration and invasion in the oral squamous cancer cell lines. MATERIALS AND METHODS: We used oral squamous cancer cell lines (SCC1483, MSKQLL1) and different gases (N₂, He, and Ar). To investigate the mechanism of plasma treatment, using different gases (N₂, He, and Ar) which induces anti-migration and anti-invasion properties, we performed wound healing assay, invasion assay and gelatin zymography. RESULTS: The results showed that NTP inhibits cancer cell migration and invasion of oral squamous cancer cell. In addition, focal adhesion kinase expression and matrix metalloproteinase-2/9 activity were also inhibited. CONCLUSION: The suppression of cancer cell invasion by NTP varied depending on the type of gas. Comparison of the three gases revealed that N₂ NTP inhibited cell migration and invasion most potently via decreased expression of focal adhesion kinase and matrix metalloproteinase activity.

PURPOSE: To investigate the effect of non-thermal atmospheric pressure plasma (NTAPP) treatment on shear bond strength (SBS) between resin cement and colored zirconia made with metal chlorides. MATERIALS AND METHODS: 60 zirconia specimens were divided into 3 groups using coloring liquid. Each group was divided again into 2 sub-groups using plasma treatment; the experimental group was treated with plasma, and the control group was untreated. The sub-groups were: N (non-colored), C (0.1 wt% aqueous chromium chloride solution), M (0.1 wt% aqueous molybdenum chloride solution), NP (non-colored with plasma), CP (0.1 wt% aqueous chromium chloride solution with plasma), and MP (0.1 wt% aqueous molybdenum chloride solution with plasma). Composite resin cylinders were bonded to zirconia specimens with MDP-based resin cement, and SBS was measured using a universal testing machine. All data was analyzed statistically using a 2-way ANOVA test and a Tukey test. RESULTS: SBS significantly increased when specimens were treated with NTAPP regardless of coloring (P<.001). Colored zirconia containing molybdenum showed the highest value of SBS, regardless of NTAPP. The molybdenum group showed the highest SBS, whereas the chromium group showed the lowest. CONCLUSION: NTAPP may increase the SBS of colored zirconia and resin cement. The NTAPP effect on SBS is not influenced by the presence of zirconia coloring.

Introdução: O tratamento de plasma é uma tecnologia eficaz que pode manter as propriedades internas dos materiais inalteradas após o tratamento, modificando apenas a superfície. Objetivo: Avaliar o efeito do plasma de oxigênio na dentina previamente exposta ao NaOCl 6%. Material e método: Foram utilizados 60 incisivos bovinos. A coroa foi removida, a raiz foi dividida e as faces planificadas, totalizando 120 segmentos referentes ao terço cervical. As amostras foram divididas em dois grupos: controle (imersa em NaOCl 6%, lavada com água destilada, seca, imersa em EDTA 17%, lavada e seca) e plasma de oxigênio (após tratamento descrito no grupo controle, plasma de oxigênio foi aplicado por 30 s). As amostras foram avaliadas qualitativamente em relação à topografia por microscopia eletrônica de varredura, utilizando-se microfotografias com ampliação de 1.000×. O goniômetro Ramé-hart foi utilizado para a mensuração do ângulo de contato entre as superfícies e as seguintes soluções foram utilizadas: água, etilenoglicol e di-iodometano. Em seguida, a energia de superfície, representada pelas componentes polar e dispersiva, foi calculada. Avaliou-se também o escoamento dos cimentos Pulp Canal Sealer EWT (PCS) e Real Sal SE (RS) na superfície dentinária. Os dados foram analisados estatisticamente utilizando os testes Kruskal-Wallis e Mann-Whitney U (p<0,05). Resultado: O tratamento com plasma levou à formação de uma camada semelhante à smear layer na superfície dentinária. Este tratamento levou a um aumento da energia de superfície e da componente polar, favorecendo a hidrofilicidade da superfície. Entretanto, desfavoreceu o escoamento do cimento PCS e não influenciou no escoamento do cimento RS. Conclusão: O plasma de oxigênio ocasionou mudanças topográficas na superfície dentinária, favorecendo a hidrofilicidade desta. Contudo, não favoreceu o escoamento dos cimentos endodônticos na dentina.


Introduction: Plasma treatment is an effective technology since the internal properties of the material is kept unchanged after treatment, modifying only the surface. Objective: To evaluate the effect of oxygen plasma on dentin previously exposed to 6% NaOCl. Material and method: 60 bovine incisors were used. The crown was removed, the root splited and the faces planned amounting 120 segments related to the cervical third. The samples were divided into 2 groups, control (immersed in 6% NaOCl, washed with distilled water, dried, immersed in 17% EDTA, washed and dried) and oxygen plasma (after treatment described in the control group, oxygen plasma was applied for 30s). The samples were evaluated qualitatively in relation to topography by scanning electron microscopy using photomicrographs at 1000× of magnification. The Ramé-hart goniometer was used to measure the contact angle between the surfaces and the following solutions: water, ethyleneglycol, and diiodomethane. Then, surface energy, polar and dispersive components, was calculated. Additionally, it was evaluated the flow of Pulp Canal Sealer EWT (PCS) and Real Salt SE (RS) sealers on dentin surface. Data were statistically analyzed using Kruskal-Wallis e Mann-Whitney U (p<0.05). Result: Plasma treatment caused topographical changes on dentin surface. This treatment led to an increase in surface energy and polar component, favoring the hydrophilicity of the surface. However, it disfavors the wettability of PCS and did not influence the RS wettability. Conclusion: The oxygen plasma caused topographical changes on dentin surface, favoring its hydrophilicity. However, it did not favor the sealers wettability on dentin.

Abstract Studies have been showing a decrease of bond strength in dentin treated with sodium hypochlorite (NaOCl). The aim of this study was to evaluate the effect of non-thermal argon plasma on the bond strength of a self-etch adhesive system to dentin exposed to NaOCl. Thirty-two flat dentin surfaces of bovine incisors were immersed in 2.5% NaOCl for 30 min to simulate the irrigation step during endodontic treatment. The specimens were divided into four groups (n=8), according to the surface treatment: Control (without plasma treatment), AR15 (argon plasma for 15 s), AR30 (argon plasma for 30 s) and AR45 (argon plasma for 45 s). For microtensile bond strength test, 5 specimens were used per group. In each group, the specimens were hybridized with a self-etch adhesive system (Clearfil SE Bond) and resin composite buildups were constructed. After 48 h of water storage, specimens were sectioned into sticks (5 per tooth, 25 per group) and subjected to microtensile bond strength test (μTBS) until failure, evaluating failure mode. Three specimens per group were analyzed under FTIR spectroscopy to verify the chemical modifications produced in dentin. μTBS data were analyzed using ANOVA and Tamhane tests (p<0.05). AR30 showed the highest μTBS (20.86±9.0). AR15 (13.81±6.4) and AR45 (11.51±6.8) were statistically similar to control (13.67±8.1). FTIR spectroscopy showed that argon plasma treatment produced chemical modifications in dentin. In conclusion, non-thermal argon plasma treatment for 30 s produced chemical changes in dentin and improved the μTBs of Clearfil SE Bond to NaOCl-treated dentin.


Resumo Estudos vêm demonstrando uma diminuição na resistência adesiva em dentina tratada com hipoclorito de sódio (NaOCl). O objetivo desse estudo foi avaliar o efeito do plasma de argônio não-térmico na resistência de união de um sistema adesivo autocondicionante à dentina exposta ao NaOCl. Trinta e duas superfícies dentinárias lisas de incisivos bovinos foram imersas em NaOCl a 2,5% por 30 min para simular o passo de irrigação durante o tratamento endodôntico. Os espécimes foram divididos em 4 grupos (n=8), de acordo com o tratamento de superfície: Controle (sem tratamento de plasma), AR15 (plasma de argônio por 15 s), AR30 (plasma de argônio por 30 s) e AR45 (plasma de argônio por 45 s). Para teste de resistência de união por microtração, cinco espécimes foram utilizadas por grupo. Em cada grupo, os espécimes foram hibridizados com um sistema adesivo autocondicionante (Clearfil SE Bond, Kuraray) e blocos de resina composta foram construídos. Após 48 h de armazenamento em água, os espécimes foram seccionados em palitos (5 por dente - 25 por grupo) e submetidos ao teste de resistência de união por microtração (μTBS) até a fratura, avaliando o padrão de fratura. Três amostras por grupo foram analisadas sob espectroscopia por FTIR para verificar as modificações químicas produzidas pelos tratamentos na dentina. Os dados de microtração foram avaliados estatisticamente utilizando os testes de ANOVA e Tamhane (p<0,05). AR30 apresentou o maior μTBS (20,86±9,0). AR15 (13,81±6,4) e AR45 (11,51±6,8) foram estatisticamente semelhantes ao controle (13,67±8,1). A espectroscopia por FTIR mostrou que o tratamento de plasma produziu modificações químicas na dentina. Como conclusão, o tratamento de plasma de argônio não-térmico por 30 s produziu alterações químicas na dentina e melhorou o μTBS do Clearfil SE Bond à dentina tratada com NaOCl.

Abstract The aim of this study was to evaluate the effect of argon plasma on dentin surface after use of 6% NaOCl. Sixty bovine incisors had their crowns removed, the roots split, and the segments planed. One hundred twenty segments of the cervical third were used. The samples were divided in two groups (n=60): Control group: immersed in 6% NaOCl, washed, dried and then immersed in 17% EDTA, washed and dried and Argon group: after treatment described for the Control group, non-thermal argon plasma was applied for 30 s. Ten samples were evaluated by scanning electron microscopy in each group. Other ten samples were analyzed by Fourier transform infrared spectroscopy (FTIR). Thirty samples were analyzed with a goniometer to measure the contact angle between the dentin surfaces and solutions, to determine the surface free energy. The last ten samples were used to evaluate the wettability of AH Plus sealer. Data were statistically analyzed using Kruskal Wallis and Mann-Whitney tests (p<0.05). The results of this study showed that argon plasma did not modify the surface topography. FTIR analysis showed chemical modifications after plasma treatment. Argon plasma increased the surface free energy of dentin and AH Plus wettability. In conclusion, argon plasma treatment modified chemically the dentin surface. This treatment increased the surface free energy and wettability of an epoxy resin root canal sealer, favoring its bonding to dentin surfaces.


Resumo O objetivo do presente estudo foi avaliar o efeito do plasma de argônio na superfície dentinária após o uso de NaOCl 6%. As coroas de 60 incisivos bovinos foram removidas, as raízes clivadas e os segmentos planificados. Cento e vinte segmentos referentes ao terço cervical foram utilizados. As amostras foram divididas em dois grupos (n=60): Grupo Controle: imersos em NaOCl 6%, lavados, secos, imersos em EDTA 17%, lavados e secos e Grupo Argônio: após o tratamento descrito no grupo controle, foi aplicado plasma de argônio não térmico por 30 s. Em cada grupo, 10 amostras foram avaliadas por microscopia eletrônica de varredura. Outras dez amostras foram analisadas por espectroscopia no infravermelho por transformada de Fourier (FTIR). Trinta amostras foram analisadas com um goniômetro para medir o ângulo de contato entre a superfície dentinária e as soluções e determinar a energia livre de superfície. As últimas dez amostras foram utilizadas para avaliar a molhabilidade do cimento AH Plus. Os dados foram analisados estatisticamente usando os testes de Kruskal Wallis e Mann-Whitney (p<0,05). Os resultados do estudo mostraram que o plasma de argônio não modificou a topografia de superfície. A análise por FTIR mostrou modificações químicas após o tratamento de plasma. O plasma de argônio aumentou a energia livre da superfície dentinária e a molhabilidade do cimento AH Plus. Conclusão, o tratamento com plasma de argônio modificou quimicamente a superfície dentinária. Este tratamento aumentou a energia livre de superfície e a molhabilidade de um cimento endodôntico à base de resina epóxi, favorecendo as características adesivas da superfície dentinária.

ABSTRACT

O plasma de baixa temperatura (PBT) tem tido seu uso investigado na destruição dosbiofilmes orais, no entanto,seu efeito sobre os substratos dentários ainda não estácompletamente esclarecido. O objetivo deste trabalho foi avaliar o efeito do PBT na durezasuperficial, no ângulo de contato, na morfologia das superfícies e composição química doesmalte e dentina bovinos. Para tanto, foram analisados a dureza de superfície Knoop (DS), asalterações nos ângulos de contato (AC), a morfologia de superfície por MicroscopiaEletrônica de Varredura (MEV) e sua composição química através da EspectofotometriaRaman (ER). Inicialmente, foram confeccionados blocos de esmalte e dentina com dimensõesde 4x4x2 mm preparados a partir de dentes bovinos. Todos os blocos foram polidos e dividosem três grupos (n=6): Plasma de Argônio, Gás de Argônio e Controle. A MS foi avaliadaantes e depois de cada tratamento, realizando-se cinco penetrações em cada espécime (50g/5spara esmalte e 25g/5s para dentina). A determinação do AC foi realizada dispersando umagota de água sobre a superfície tratada e observando o ângulo de contato obtido em 1 min e 1hora, utilizando fotografias digitais de alta resolução e o programa Image J. As alteraçõesfísicas na morfologia do esmalte e dentina submetidos ao PBT foram observadas a partir daMEV. Adicionalmente, a proporção relativa do conteúdo orgânico e mineral do esmalte edentina após os tratamentos foram analisados por ER. . Os dados foram avaliados por análisede variância (ANOVA) – 1 via para MS e 2 vias (tratamento e tempo) para AC, após Tukeyteste (α < 0,05). Os resultados da MS demostraram não haver diferença significativa noesmalte (p = 0,087) após os tratamentos. No entanto, a MS da dentina diminuiu após otratamento de plasma de argônio (p = 0,005)...


Low-temperature plasma (LTP) is a promising technology with the potential benefit ofdestroying the matrix of the oral biofilm and also produce antimicrobial effect on the bacterialcell. However, the effect of LTP on dental structures is not clear. The objective of this studywas to evaluate the effect of LTP on enamel and dentin substrates. Therefore, we analyzedKnoop microhardness to calculate the percentage of surface hardness (SH), changes in contactangles (CA), the surface topography by Scanning Electron Microscopy (SEM) and also thechemical composition by Raman spectrometry (RS). Initially, 4x4x2 mm enamel and dentinblocks were prepared from bovine incisors. All blocks were polished and divided into threegroups (n = 6); argon plasma, argon flow rate and control. The SH was assessed before andafter each treatment, performing five penetrations in each specimen (50g / 25g and 5s enamel/ dentin 5s). Contact angles changes were performed by dispersing water drop on the treatedsurfaces and observing the contact angle obtained after 1 min and 1 hour. For that, highresolutiondigital photos and the Image J program was used. Physical changes in the structureof enamel and dentin submitted to LTP were observed from the SEM. Additionally, therelative proportion of the organic and mineral content of the enamel and dentin after treatmentwere analyzed by RS. Data were evaluated by analysis of variance (ANOVA) - 2-way(treatment and time) and Tukey test (α <0.05). For the SH the results showed no significantdifference in the enamel (p=0.087), however, the SH of dentin decreased after treatment withargon plasma (p=0.005)...

Propriedades adesivas dos materiais odontológicos, como molhamento e rugos idade de superfície são importantes para estudos de efeitos de cimentações de peças protéticas, restaurações e reparos, unindo diferentes materiais e, até mesmo,adesão de células em implantes dentais. Modificações nas superfícies desses materiais podem contribuir para melhoria dessas propriedades. Este estudo analisou os efeitos do tratamento com plasma não térmico em superfícies de titânio puro,cerâmicas vítreas reforçadas por dissilicato de lítio e cerâmica feldspática. Espécimes de titânio foram distribuídos em três grupos (n=5): sem tratamento (ST);tratamento com gás argônio, 6 minutos; e tratamento com plasma de argônio, 6minutos, 10W/1 bar. Foram analisados: ângulo de contato logo após o tratamento(TI), após uma hora (TH) e após uma semana (TS); rugosidade de superfície (Sa); etopografia de superfície (MEV). Espécimes das cerâmicas foram distribuídos em dois grupos (n=5): ST e plasma de argônio 6 minutos, 10W/1bar. Adhesive properties of dental materials such as wetting and surface roughness areimportant for studies of cementing effects of prosthetic devices, repairs andrestorations, combining different materials and even cell adhesion in surgicalbiomaterials. Modifications in the surfaces of these materials can contribute toimprove these properties. This study examined the effects of treatment with argonplasma on surfaces of pure titanium, glass-ceramics reinforced by lithium disilicateand feldspar ceramic.
